چکیده

In previous studies we found that a commercial cholesterol preparation obtained from the Wilson Laboratories had a high concentration of antirachitic provitamin. In the present studies we undertook to concentrate the provitamin fraction in cholesterol and to study its properties. 175 gm. of the commercial product, Cholesterol W, were dissolved in 1200 cc. of 10 per cent solution of sodium hydroxide in alcohol and boiled under a reflux condenser for 1 hour. After chilling the solution to 38”, the mass of crystals was drained free from liquid by suction and washed with cold alcohol. The alkaline alcoholic filtrate was evaporated to dryness and extracted with boiling acetone. After evaporation of the acetone, the brown, semicrystalline mass was taken up in ether, the ether solution washed repeatedly with water and finally evaporated to dryness. The residue was still light brown and somewhat waxy in consistency. This was Fraction lc. The large mass of crystals separated from the alkaline alcohol above was dissolved in ether and this ether solution washed by shaking with water until the wash water was only faintly alkaline. A deep emulsion layer ‘containing cholesterol always formed between the water and ether layers. This was separated, the cholesterol filtered off, dried, and kept as Fraction II. 157 gm. of cholesterol obtained from the original ether solution were recrystallized twice from acetone. 113 gm. were recovered. This was Cholesterol B. 50 gm. of Cholesterol B were recrystallized from acetone three additional times with a final recovery of

Nature of the provitamin D of the ribbed mussel, Modiolus demissus (Dillwyn).

Since 1934, when it was shown that ergosterol is not the only provitamin D occurring in nature (l), the provitamin D content of the mixed sterols from many sources has been investigated (2-6). It has been found that the sterols from many of the invertebrates contain a surprisingly large amount of provitamin D as measured spectroscopically. Windaus (3) and Bock and Wetter (4) were able to isolat...

Natural Abundance of Serum Retinol Is a Novel Biomarker for Evaluating Provitamin A Carotenoid-Biofortified Maize Consumption in Male Mongolian Gerbils

Background: Crops such as maize, sorghum, and millet are being biofortified with provitamin A carotenoids to ensure adequate vitamin A (VA) intakes. VA assessment can be challenging because serum retinol concentrations are homeostatically controlled and more sensitive techniques are resource-intensive. Objectives: We investigated changes in serum retinol relative differences of isotope amount r...

CD36 and SR-BI are involved in cellular uptake of provitamin A carotenoids by Caco-2 and HEK cells, and some of their genetic variants are associated with plasma concentrations of these micronutrients in humans.

Scavenger receptor class B type I (SR-BI) and cluster determinant 36 (CD36) have been involved in cellular uptake of some provitamin A carotenoids. However, data are incomplete (e.g., there are no data on α-carotene), and it is not known whether genetic variants in their encoding genes can affect provitamin A carotenoid status. The objectives were 1) to assess the involvement of these scavenger...

Bioconversion of dietary provitamin A carotenoids to vitamin A in humans.

Recent progress in the measurement of the bioconversion of dietary provitamin A carotenoids to vitamin A is reviewed in this article. Methods to assess the bioavailability and bioconversion of provitamin A carotenoids have advanced significantly in the past 10 y, specifically through the use of stable isotope methodology, which includes the use of labeled plant foods. The effects of the food ma...
